The duomo is not a museum, it is an operating church, therefore it is always open except at night. Usually there are masses and other rites going on since early morning (first mass is at 7.00) until late evening (last mass is at 18.00). The ascent to the roof, though, is possibile only since 9.00 to 5.45.

You can purchase tickets to take the elevator (aschensore) to the roof, which if you go on a clear day is a fantastic view, but a bit of a walk even with the elevator. As stated below, going inside the Duomo is free - women make sure to have their shoulders covered or they will not allowed inside. No shorts for women.
