We are going to be in Florence on the 1st of May and then hoping to travel to Rome by train on Monday 2nd May can anyone tell me what sort of things will be closed because of the Labour Day holiday? Thanks for any help.

All state-owned museums like the Uffizi, Accademia, Bargello, Medici Chapel etc will be closed. Churches will be open but attached museums may not.
See http://www.uffizi.firenze.it/english/orari/orari.asp
Shops closed, bar and restaurants mostly open.
As May 1st is a Sunday, slight variations are possible.

Italy doesn't transfer a holiday falling on a Sunday so should be open. Some shops close all day Monday or open only in the afternoon.
