We did that drive a couple of years ago. The directions we followed said 160 miles should take about 2 1/2 hours. However, we stopped for lunch and ran into some fog and a little bit of traffic so it took about 4 hours.
We found that whatever directions we were given, the driving times were always longer, usually by about 20%...sometimes longer depending on how lost we were.
By train on the EuroStar, the trip is about 3 hours.

From Venice to Florence is normally 3 hours to drive. Take A4 to Padova then A13 to Bologna and then A1 to Florence. One way tolls will be 18 euro. Try to time your drive so that you are not passing Bologna between 0730 to 0900 or 1700 to 1900 this is the highest traffic time.
There are trains all day and if you are not in a hurry this is the least stressful way to go.
