Holland Tunnel to The Roosevelt Hotel
I will be arriving Sunday morning via the Holland Tunnel and would like to go north on The Avenue of the Americas to show the family the city. I notice there are a lot of bus only lanes where I want to turn right. I want to turn right onto E. 40th then left onto Madison Avenue. It is on Madison Ave. that I am seeing two lanes of bus only. I want to turn right on E 44th. Is it legal to do so?
My objective is to get to the Roosevelt Hotel by turning right on 40th then left on Madison Ave.,right on to E 44th, left onto Vanderbilt Avenue, left onto E. 45th to the doors of the hotel.
I have been using Google Maps street view to see how intersections are laid out.

Bus lanes means buses only for driving - if you need to turn you can move into the bus lane to do so but only right before the turn.
However, I know that in that area there are some turns that are illegal and I'm not sure exactly which intersections (I know many from 42nd St).
I would call the hotel - concierge can tell you - to find out if there are any turn restrictions and what days and hours they apply (many are only during rush hours).

If you are going to the Roosevelt Hotel, I'd actually recommend exiting the Lincoln Tunnel and make your way to 38th Street. Turn left on 8th Avenue instead of 6th (it's a more interesting drive and you see Broadway theaters that way). Then turn right onto 48th Street and drive straight through to 5th Avenue and turn right, then turn left onto 44th, then left on Vanderbilt, left on 45th, and straight to the hotel.
It should be relatively quiet if you enter Manhattan early enough on a Sunday morning, and while this is a little roundabout, you'll see the Times Square area and Rockefeller Center. I can't remember if you can enter the hotel directly from Madison, but you may find that there's an entrance there.
