I don't know if you can catch a bus from Boston to Falmouth -- but, check with Brush Hill and the Plymouth & Brockton lines. But, as an alternative, you can take the bus from South Station to Hyannis, and take the ferry from Hyannis to Martha's Vineyard.

Depends where you're staying.... I always rent a bike down in Edgartown along the waterfront. A great ride is along the ocean between Edgartown and OB. Not far, but so lovely! Lock you bike up in OB, grab a burger on the boardwalk and continue on to Vineyard Haven for icecream at Mad Martha's! Work it all off on the bike ride back to Edgardtown!
I'd stay away from the moped option - too many cars and other mopeds in the summertime, though it is a nice way to get out to the Cliffs if you're up for the adventure!
