From the Museum, you are on the West side and Grand Central is on the east side, so you will have to come downtown and then head east.
Probably the easiest is to take the B train right under the museum down to 42nd St (and 6th Ave) and walk about 4 blocks over to Grand Central. The A train does not stop at the Museum, it is an express train.
The B is a local train and will take you a couple of blocks east than the C train which also stops at the Museum.
Have a fun time at the museum.

One way or another, this trip will involve walking a few blocks, so you just have to decide how you want to do that walk. The easiest solution (to me at least) is to exit the museum on the south side and then walk west to the 1 train at 79th street (5 to 10 minutes depending on how fast you walk). Then take the 1 to Times Square and transfer to the S shuttle to Grand Central. These are the most frequent trains.
The other directions also work, though I'd avoid the C, which doesn't run very often and takes you to the far west side, requiring a much longer walk. The B train is a better alternative, though still not what I'd choose.

chestnut, hopstop.com is an excellent resource for getting around NYC. For any given search, you can tell it if you'd like to do more walking/less transfers or vice versa. And if you put in the date/time you are expecting to go, the site takes into account any construction going on, traffic at a particular time of day, etc.
That said, for this particular journey, I agree that the *B* to 42nd Street or the 1 to the Shuttle are your best bets. You do *not* want to have to transfer from the A/C to the Shuttle (or the 7) at 42nd Street because it's a *very* long and convoluted walk underground. (The A/C is underground at 8th Ave; the Shuttle is at the other, eastern side of a very large transit station.)
FWIW, hopstop would have directed to take the A on a weekend (e.g., yesterday when sf7307 input it) if there were service changes due to construction; it often runs local on the weekends.

I've used google maps as well as hopstop to get directions for city travel. We've taken the B train to 42nd and walked a short way to GCT. If you get concerned at all, don't hesitate to Ask For Help! New Yorkers are always willing to help visitors navigate the mass transit system.
