DAY TRIP TO CINQUE TERRE FROM FLORENCE?

Day Trip (singular) or Trips (plural)? If you want to go more than one day, I'd stay over in the Cinque Terre itself somewhere.
Look at the Trenitalia website to check train connections from various cities like Florence and Pisa. You might find a few direct trains a day from Pisa or even Florence; otherwise, expect to change at La Spezia most likely. Then you can decide if you wish to spend that much time on a train for a day trip.

Florence to one of the towns in the CT is 2.5 hours each way, so it's 'possible.' Depending on the time of year, the towns could be packed with tourists. Whether that's the best use of your sightseeing time is your call.

We drove from Lucca and had most of the day. Leaving early, we arrived back at the villa at 8pm, but we had a chef with dinner waiting.
There were 12 of us and 7 of us walked four towns, about 6 miles along the path above the towns.
We parked at the first town and took the boat to the 4th town and walked back. Five took the train back.
Not sure now if the trails are completely walkable after the floods.

You are looking at 6 hours on trains and 4 trips going back and forth from stations. The villages are about 15 minutes apart. Expect crowds.

We drove from Florence to the Cinque Terre. We parked at the La Spezia train station and took the train to Riomaggiore. We went to all five towns including two by hiking trails. We made the easy hike to Manarola, took the train to Vernazza where we had a very good lunch at Gambero Rosso, made the more strenuous hike back to Corniglia, then took the train to Monteroso al Mare. We left early in the morning and returned late at night but we are glad we went and recommend it highly. Of course it would be better to spend a few days there but we will have to do that another time.
