Take a taxi. It's the quickest and easiest. It will cost somewhere around $40. The roads are fine. The snow is already quite a bit melted and they did a good job plowing. The likelihood of having another snowfall of any consequence is next to nothing.
You will hit some rush hour traffic, but you hotel is not far from the midtown tunnel (most likely the route into Manhattan).

If $ are more important than money I think you can get the hotel "direct" to your hotel for about $20. The only thing is - the shuttle goes direct to a lot of hotels - and since you're pretty far downtown it may take quite a while (1.5 - 2 hours) for the trip - depending on how many hotels they stop at further uptown.

Taxi or car service. Cost is about the same (the car service can be less if heavy traffic).
At 4pm, I'd probably reserve a car because the taxi line could be long.
Also, if it matters, you can pay a car service with credit card.

If you're 1 person, not in a rush, take the Airport Shuttle that goes to PENN STATION which isn't far from your hotel. If there are 2 of you or you're in a rush, take a taxi or order a car service from
