Clonard -- I suggest you get a good guidebook or surf the New York info on this site. New York is a big city with lots of attractions. There is no hotel that is close to all of them and yet, with its easy to use and reliable public transportation system, there is really no hotel that is "far" from anything you will want to see either.
Search the posts here for hotels and you will see recommendations for some reasonably priced hotels.

Here's a one bedroom apartment that sleeps 4 for $225 a night (note that without taxes that is equal to the price of a hotel listed at about $190 per night).
It's in the world Plaza on 50th between 8th and 9th, subway adjacent, great views, doorman. I clicked the calendar and other than the first few nights in Sept., it is available.
We're renting it for 9 nights in May.

Please be aware that September is heading into the busier season in NYC and you may find that prices then are higher - perhaps significantly - than in March or April.
Also - tourist sites, nightife etc are located all over Manhattan - and even excluding outliers you're talking an area 8 miles long and perhaps 2 miles wide. No one pllace is close - or even really walkable at all to every thing.
I sugest you:
1) Read the posts above on NYC sitesa dn hotels
2) Come back and give us more specific info - what your hotel budget is and what things are most important to see (or what neighborhood you like the sound of) - and this may differ if you're talking 2 couples or if there are any kids involved
