Budget? Interests?
Without knowing your budget or interests, here are some general suggestions:
1. Take a Gullah tour
2. Visit Patriot's Point
3. Have a drink on top of the Vendue or Marketplace Pavilion
4. Ride out to Shem's Creek for lunch on the dock
5. Walk the length of King St.
6. Stop in the Charleston Historical Society and inquire about garden tours.
7. Walk the Battery

Go Travel did a fabulous 'trip report' to Charleston with tons of great information, also will come up a bunch of other threads , trip reports, restaurants, sites, from those who posted from recent trips -
have fun ! I thoroughly enjoyed my visit last year

If you are willing to drive southwest for about 25 minutes, you will find a true gem for good, country eats at Bubba-Cue's Bar B-Cue House, 6165 Savannah Hwy., Ravenel, SC. Bubba-Cue's has tender, moist and still flavorful pulled pork, succulent friend chicken, the best macaroni and cheese my daughters claim to have ever eaten and delicate, sweet hushpuppies that stopped me from crying.

I would highly recommend going to the visitors center and then taking one of the mini bus tours. The busses are located behind the visitors center. We had avoided doing this for years, and then we took one of the tours. It was wonderful.
I would also recommend California Dreaming for a meal.
