A car would be best, but it is possible to get there by bus/train combo. From Florence you could take the train to Siena (about 7€). You would need to change to a Rama Mobilita bus from Siena to Pitigliano. While there are lots of trains between Florence and Siena, there are few bus departures on to Pitigliano.

There are excellent bus connections with RAMA (Florence/Grosseto train station; Grosseto train station/Pitigliano:

I think that you'll find the routing via Grosseto to be more convenient than the routing through Siena.

I only see three buses total for Grosseto/Pitigliano, only two of which would work with connecting from Florence via Grosseto as the other departs too early.
You would only have one bus connection from Siena or two from Grosseto.
