Cheapest is the TTC. Bus from Pearson to Kipling Station then subway by Bloor and Yonge lines to College Station. You should be able to walk to your hotel on Carleton from there. Cost is $3. Time is about 45 minutes.
How do you define "best"? Fastest? Most comfortable? Least hassle? There are many choices.

The Union Pearson Express rail link between the airport and downtown is now up and running. With only two intermediate stops, the train should avoid the almost inevitable Toronto traffic jams. Estimated travel time of around half an hour. It should also be more comfortable. But not cheap. Full fare is $27.50 with discounts for younger riders and seniors. Tickets are available on-line. The downtown terminus is on the west side of Union Station. http://www.metrolinx.com/en/projects...upexpress.aspx
