Is Mestre the easiest place to pick up a rental car in Venice? We will be staying in the Dorsoduro area, if that matters. And in Florence, it seems dropping off at the airport is the best bet to avoid the ZTL, correct?

I picked up at P. Roma in Venice and dropped off at Via Borgo Ognissanti in Florence, which is not in the ZTL and is easy to get to.

P Roma is straightforward - boom you're on the causeway going out - Mestre you'd have to take a shuttle train to. Florence rifredi train station, a few miles out of town may be an option for Florence is joannyc;s doesn't suit you - trams go from Rifredi to the center of Florence. Rifredi is on the north side of Florence coming in.

Most rentals have agencies in Borgo Ognissanti. Be careful as higher number of that street are not in the ZTL, while lower numbers are.

Yes that Borgo Ognissanti area is easy to reach without a lot of traffic or small streets - the American Consulate is there - and it at least along the river has broad streets. Seems easy to me by looking at maps.
