I think it would be difficult to do day tours from Colombo or Galle. Most of the parks and the Cultural Triangle are located on the opposite side of the island. I would suggest staying in the Cultural Triangle and doing day trips from there.
Wildlife probably means Yala. Not sure what you mean by "unique" - by definition if it's unique none of us have done it already.
Jreinho is asking for "unique cultural and wildlife tours" in many places there days, this is the third one I've seen.
If you are interested in a combination of culture and wildife in Sri Lanka, you need to stay in the cultural triangle area or the Yala area.
