You can see the hours at both the Times Square and Downtown locations at the following link:

My most recent experience in waiting for Saturday matinee tickets at the Times Square location was about one month ago. I arrived at 10:00am and waited about 1 1/2 hours to get to the window. I was able to get tickets for the show that I wanted (42 Street) with no problem. Given the number of people in line ahead of me at 10:00, I would think that you would need to arrive at least an hour before the booth opens to be anywhere near the front of the line.

Yikes! I can't imagine waiting in line this long for show tickets...do you save that much to make it worth while?? Do you get a better deal there for last minute tickets than say on ebay.com? I'm planning to visit NYC in October and would love to see a musical or two. Thanks.

For matinees, it's best to get in line before 9:30 a.m. It opens at 10.
For evening performances, get in line a little after 2.
To help you figure how long a wait: Each blocklong length of the line (yes, it can snake the equivalent of 4-5 blocks) takes about 20-30 minutes waiting time until you get to buy your tickets.

Which location makes the most sense really depends on your plans for the day. Going downtown for tickets may not save you much time if your plans for the day are in midtown. I would say choose your location based around your other plans for the day.
