Key Biscayne is very close to Miami and is a beautiful area to visit, especially down by the lighthouse.
Key West is wonderful. It's a good 3 hour drive from Miami, so I would definatly spend the night. Although I haven't been to these attractions, I've heard great things about the Key West trolley tours and the Hemingway House.

I suggest you get a copy of Joy Williams guide to the Florida Keys. It will give you a very good idea of what the different Keys are like.

It is a beautiful drive from Miami to Key West. Stop at Craig's restaurant (Mile Marker 90) on Plantation Key for the best fish sandwich in the keys!
Definitely spend the night in Key West.
Go to Mallory Square for the sunset celebration. See the "Cat Man" show on the Sheraton pier.Take the Conch train tour or the Trolly tour to see the whole island.
Go to Fort Zachary Taylor for the best beach in KW. There is a small fee but well worth it.
