For how many people? what kind of luggage?
Taxi is the easiest, door to door , but the most expensive. Car services cost about the same.
For one person without major luggage, a special bus (not the regular city bus,) to midtown is a possibility. Then you have to walk a few blocks to your hotel.
For 2 or 3 people, just take the taxi from the airport.
Regular bus to subway options are cheap, but take much more time and involve dragging suitcases on an off and up and down stairs

I fly in/out of LGA several times every month.
If you're landing at rush hour or during the holidays when it's always busy, reserve a car service because the taxi line can be quite long.
Otherwise, simply take a cab.
(Public transportation options are horrible, unfortunately, and the "super" shuttle service receives awful reviews. Unless you're truly destitute, take a taxi or car service.)

You can take an Airport Express bus to Port Authority for $16 (I think) and then walk several blocks to your hotel - but sidewalks will be very crowded. This is cheaper than cab for one person.
For 2 or more people cab won;t cost any more.
There are couple of true public transit options (bus to subway) but they will take 1.5 to 2 hours and you will have to haul your luggage up and down stairs and down long passageways. (Go the the LGA website for the 2 options.) (I wouldn;t do this is you paid me - luggage on the subway packed with commuters is NOT fun.)
