What does "splurge" mean to you?
If you are willing to pay upwards of $15,000 to $20,000/week for mid-September there are some really very nice beaches houses you can rent. Do a google search for "Southampton Villa Rentals" and you'll find lots of listing in many price ranges.

In east hampton, the nicest places are the Baker House Inn, The Mill Street Inn and teh 1770 house. each of these have a wesbite so you can look at the rooms and the property. You can also search for some posts I started asking Hamptons lodging questions.

You need to realize that the towns aren't on the beach - and that you will need a rental car - plus a town sticker from your inn - in order to park at the beach (except for Hither Hills state park).
As for towns, Southhampton is uber wealthy and uber trendy (lots of Hollywood people) East Hampton is uber wealthy - but more artsy/somewhat less ostentatious (more writers etc).
Neither will be cheap - I like East Hampton better for gallerys, small local museums and cute (non-chain) shops. (Southhampton has all the designer shops - as in the window saying London, Paris, New York, Southhampton and Nice or Monte Carlo.)

Mid Sept. is usually still quite beautiful, sometimes warm Indian summer, sometimes start of fall. If you will have a car (good idea) it doesn't really matter whether you stay in Southampton, Bridehampton, Wainscott, East Hampton or Amagansett imo. Pick the town that has the kind of place you want (inn, b & b, cottage, motel-in town or on the beach) and hope you can find it available and at a price that suits you.
If you want to be able to walk to restaurants, staying in Southampton or East Hampton village is best. emcash has given you the names of the nicest EH places. In Southampton, A Butler's Manor gets very high ratings on tripadvisor (a b & b). 1708 House is also nice.

Btw, I don't agree with the descriptions of the towns above. East Hampton has more celebs/Hollywood folks and is a LOT more ostentatious and I suppose trendy (think Tiffany, Gucci, Coach etc.) than Southampton which has historically been more old money. I prefer the restaurant options in East Hampton, but if you have a car, it's not that far between any of these towns.

As a local year rounder I have to say that Sag Harbor is the BEST town of the Hamptons. We sport our celebrities but in a much quieter way than the other Hamptons – I won’t even tell you where Billy Joel lives But you can look at his boats in the harbor. If you want a real posh experience stay right in the center of town at the American Hotel. September is PERFECT especially if you come during Harborfest – so homey with a parade down Main St. and whale boat races. Take the Hampton Jitney directly to the American then rent a bike across the street at Bike Hampton so you can ride to the beach on the bayside. For the ocean beaches you can take a cab or rent a car – it’s about 9 miles. Enterprise car rental is great out there cause they ‘will pick you up”. Enjoy the east end!
