We stayed at this lovely hotel in a great central location in Munich.

You can walk to many places like the food market, the Rathaus, Deutsches Museum, a toy museum, many restaurants, or take the S-Bahn or U-Bahn. You may need to rent two rooms; I'm afraid 5 in one room is going to be difficult to find. You can arrange a bus tour to the castles from there if you wish. You get to see a lot, but it is a bit of a drag not being able to set your own schedule.

As per your other thread, you cannot put your 6year old in your bed, and the other two kids in a second double in the same room. Room occupancy is limited. The 6yesr old could sleep in a single in your room if the room occupancy states that is allowed. Don't try and smuggle the children into your room, even if it has two doubles.
You may find a family room for 5, but it is doubtful.
Look at sites like booking.com where you can fill in the size of your party to see if such rooms exist. Otherwise you need at least 2 rooms, which may not be adjoining.

The Viator tour to N'stein from Munich usurps 9 hours and probably costs around $200 for your family. The onsite tour of Neuschwanstein - which is not a castle, by the way - is the only way to visit N'stein whether you use Viator to get there or public transport. The onsite tour takes a mere 30 minutes and is pretty dull, IMO. The public transport option (which would cost far less) still requires 5 hours of train travel to get to N'stein and back.
All in all, the 30-minute tour of this faux "castle" isn't worth the the time or the money, IMO, unless you have other travel objectives in the area. There are other things to see and do around Füssen that might justify a visit there; if you decide go, I would plan to spend a couple of nights.
If you prefer to stay in Munich and do a daytrip somewhere, there are other good options that might suit you. Prien is just one hour by train east of Munich; there you can see Ludwig's Herrenchiemsee Palace, on an island in Chiemsee Lake:

The standard hotel room in europe had one double bed for 2 people. For a family room - 4 or sometimes 5 people (if there is room to add a roll away) need to be reserved as such. You can;t simply put another person in the room - against fire laws and hotel rates. Especially if breakfast is included you wold just be getting a freebie.
I would start looking at the discount hotel sites and be specific that you need a family room for 5 - 2 adults and 3 children. do not let them give you a "cot" for the 6 year old as that is British usage for an infant crib - not a camp bed.
