if you are going to be there for a week or so it may be worth it,but not by much.I travel to Prague quite often and have never used this card, from what i can gather there are several of the more popular places that do not accept it.
Have a look at this explaination.

BTW this website is a very very useful tool for someone new to Prague. The website is run by an expat who has lived in Prague for many years

It's like Venicecard that entitles you to rides to public transportation + entrances to museums (+ use of public wc in the case of venice). And from what I read on the different forums, more people seem to find it not worth. I have been to Venice and Prague roughly 10 times each in the recent past (each time a stay of 4 - 5 days) but, like unclegus, have never bought one myself. Usually I buy 24 - 72 hr pass for public transportations (tram, bus & metro in Prague) and pay the entrance to the museum each time. I don't go to that many museums at each trip so that's fine with me.

an interesting thing about the transport tickets in Prgaue is that it is more expensive for a 3 day pass than it is for 3 x 24hr passes.
The transport authority put the prices up about a year ago.
A 7 day pass used to cost 280 czk a 5 day pass now cost 500czk. ( the 7 day opass is no loger available)I think this is purely aimed at the tourist trade as the locals all use monthly passes.

this gives details.
the transport system in Prague is very good and ticket covers you fro buses trams and metro system.

Yes, I thought the price increase was outrageous (especially when you know the older prices).
3-day/72hr pass : CZK 330
1-day/24hr pass : CZK 100 x 3 > 300
Above is what unclegus means. One detail, with 3-day pass you can take one child while you cannot with 1-day pass. I'm usually alone or with another adult so it's not an advatage at all. Seems only an excuse to me to justify the fact that 3-day pass is more expensive than 1-day x 3.
