Delphi, without a doubt. It's one of the most important sites of the Greek civilization to which we owe almost all of our Western civilization.
Budman refers, probably, to the one-day trip that takes you to Hydra, Aegina and either Spetsae or whatever the other island is called. (My trip was almost 30 years ago.) Hydra is picturesque; Aegina has a lovely Greek temple.

Delphi. Absolutely breathtaking from a scenic, historical, literary, or mythological perspective.
We took a ferry to Aegina for a day. Beautiful temple, well preserved. I had considered the cruise to three islands but ended up spending much less money on the metro and ferry ticket to Aegina, and we had more time to explore the island and less time on the boat.

Nafplion deserves at least an overnight stay to enjoy its charms, and to explore the surrounding archaelogical sites (Mycenae, Epidauros, etc).
But if your schedule really only permits Delphi *OR* Nafplion , then Delfi is more deserving.
I would try to extend my trip & do both !
Hydra island makes a very nice day excursion.

I think Delphi for the one day tour. Although I like Nafplion an awful lot, agree that it requires a little more time.
Like Budman, I took the one-day cruise. It went to Aegina, Poros, and Hydra. If you are interested in the ruins, Aegina is best. But Hydra is the most charming of the nearby islands. (By the way, the '56 movie "Boy on a Dolphin" with Sophia Loren was filmed in Hydra. Its a dull movie, but nice scenery to see before you go).
