If you take the train, you should transfer at either Roma Trastevere or Rome Ostiense. You don't need to go all the way to Rome Termini. From both Ostiense and Tiburtina, there is a commuter train every 15 minutes (30 mins. on Sundays) to Fiumicino Airport. This commuter train is NOT the "Leonardo da Vinci" express non-stop airporter train from Termini.

The port and the airport are both on the coast, with the airport about 40 miles south of the port. By car, it should take less than an hour. With luggage I think a taxi or car service would be best. A train would take longer than you want and with changes and luggage could be a pain. You might want to check with some car services to get price quotes. Some recommended on this site are:
