One way would be to take the train from the airport to the Termini station and the Metro or a taxi from there to your hotel; there is a Metro station at the bottom of the Spanish Steps. The Metro tickets are best bought at a tobacco shop or newsstand since the ticket machines can be either out of order, hard to use, or both.

Take the train from the airport to the Termini train station and then either a) take a Metro (Line A, Direction Battistini) and get off at Spagna, or b) take a taxi. If you take a taxi from Termini, go the taxi rank rather then accepting an offer from a taxi driver at the Termini entrance. The metro is easy to use, but it can be crowded and uncomfortable, particularly if you have lots of luggage. You'll get more responses to future questions if you specify that you're going to Rome.
