Janev,
Amidst the various markets you've received a bit of a hodgepodge of answers, and I'll attempt to be a bit more precise to make your day travelling in Melbourne that little bit more efficient.

Thus:
Catching the 109 should be no drama as it is a case of walking down the pier and it'll be about 300M away off to your right a bit - would be surprised if there are not directional signs and a ticket machine as it is the terminal stop.
Last time I bought a Zone1[all you will need] day ticket it was $6.50 but could be $7,?? by now.

If you want to go to the St. Kilda Markets, catch a taxi along the esplanade, about 4-5 km. and then you can get a tram back into the CBD from St.Kilda - reason btw you cannot use the metlink site is there'ssome note about maps upgrade.

So one way or another you get back into the city and if it is with the 109 you could stay on it until you get to the Swanston Street Mall [you'll be at the City Square with Townhall on your left - head south or to your right a block and you're at Fed Square opposite Flinders Street Railway Staion with all the clocks, and Young and Jacksons Pub and St Pauls Cathedral on the other three corners, all famous Melbourne landmarks and over at Fed Square[weird looking buildings there] there are some steps that take you down into the Visitor Info Centre, great place for maps etc.

OK, downunder gave a metlink link for the free cuty circle tram, also mentioned by marg and it will save your legs and a lot of time - you can catch it on Flinders Street right opposite the Fed Square and they are older type trams of a Burgundy and Cream trim - you will not mistake them and they run continuously between the sated hours and will take you right around the Golden Mile and you can hop off and have a look and then get on the next one, there being Spring Street with Treasuey and Fitzroy Gardens, Captain Cooks Cottage, government buildings and churches and theatres with the Exhibition building in Carlton Gardens at top of Spring Street and behind the Exhibition Building is the new Melbourne Museum and worthwhile just to walk around behind it to see somereally interesting architecture blending with the park.

And so the tram will take you past the Old Melbourne Gaol where Ned Kelly was hanged, and then further along a re the Vic Markets and then further still it will do a loop around docklands new development area and then back along Flinders Street and you could hop off at the Fed Square again and walk past the railway station over the Yarra River and you'll see Southbank.

Do that first if you want to go to those markets.

But the City Circle tram is by far the easiest way to get around and if you have enough time or it turns out to be a hot day, plenty of relief to be found in parklands and/or Botanical Gardens heading south east or east along the south side of the Yarra river.

Melbourne is a very easy city to find your way around as the CBD is laid out on a grid system and down near the river, it is all open and airy, very easy to see what's about and with a map and the tram you can be your own guide.
