I'm only flown business class on a 747 once, but here are the usual advantages and disadvantages of getting seats upstairs:
Advantages:
- fewer passengeres in your section
- quieter
- sometimes better service because of fewer passengers
- you won't have economy passengers walking by you when boarding
- cool factor
Disadvantages:
- have to climb stairs, especially if you have large carryons
- small overhead bins. large carryons have to be stored in a cabinet by the FA
- the cabin is narrower and shorter, so it's like you're flying on a narrow-body plane, not a 747
- takes longer to deplane

I used to fly,regularly, upstairs in business class back when Pan Am was around and I had over 300,000 frequent flyer miles in my account. Back then a business class ticket to Europe, for example, only cost one 60,000 frequent flyer miles. I always preferred flying upstairs over downstairs. It's quiter and there were fewer passengers for restroom use. Plus the flight attendants had fewer people to serve upstairs. I REALLY miss those days.Happy Travels!

I've flown upstairs many times. It is quieter, no question. But to me it feels more cramped. Your regular carry on won't fit near your seat, so if you use things from your carry on that's a hassle.
Everyone has their own favorite places to sit. I prefer to be downstairs in a bulkhead seat in Business.
