We hope to visit Cape Cod in September, staying in one of the RCI timeshares for a week. Which town would be best? Falmouth (east or west), Hyannis, Provincetown, Brewster, South Yarmouth, Dennis Port? We plan to rent a car in Boston, and perhaps spend the first night in Plymouth. Is it worthwhile to see Plimouth Plantation? This will be our first trip to Cape Cod. Hopefully, we'll have time to do one-day trips to Nantucket and Martha's Vineyard.

Brewster, DennisPort t and South Yarmouth would give you a nice mid-Cape experience. I recommend a day trip to Provincetown. Plimouth Plantation is interesting and the replica of the Mayflower in in the town of Plymouth near Plymouth Rock.
How much time do you have? The Cape is large and each of the main waterfront areas- Atlantic Ocean, Nantucket Sound, and Cape Cod Bay plus myriad fresh water ponds and lakes all give you a different experience.

We'll have one week in Cape Cod. At this point, I'm thinking of staying one night in Plymouth before driving down to the Cape.

IMO, not really worth the time to overnight in Plymouth unless you're waiting for your timeshare to become available, or haven't time otherwise to visit the Plantation. Avoid Hyannis.

Hyannis does not have great beaches and it does have lots of traffic, especially on rainy days.

It is fine for a visit and a meal, but I wouldn't stay there.

In general, the west side of the Cape along Buzzards Bay, the north side of the Cape along Rte 6A, and the Turo-Eastham area are all attractive/quaint areas of the Cape, while the towns along Nantucket Sound and Provincetown are a bit more beachy.

Of your choices, I would aim for Brewster.

I don't know about staying one night in Plymouth but you should visit this historic site IMO...and other site if possible. Enjoy the Cape!
