The first one I think of is Southwest Airlines <http://www.southwest.com/>, but a lot depends on when you want to travel. Can you give us some more details?

I like to use www.kayak.com for comparison shopping, and then book my flight directly with the airline.
It does not show fares for all airlines (i believe it does not include Southwest), so also check what OV has posted.
Be sure to include other nearby airports like Fort Lauderdale (FLL).

JetBlue (from BUR Burbank and LGB Long Beach airports in Los Angeles) and Southwest (from LAX and from SNA Santa Ana/Irvine airports in/near Los Angeles)
fly to Fort Myers but neither shows up on some consolidated search sites, check their websites individually.
If you can fly into Fort Lauderdale, you can use both JetBlue (from BUR and LGB) and Southwest (from LAX, BUR, SNA as well as from ONT Ontario near Los Angeles),.

When are you flying? American currently has quite a few choices at $129 (that's $139.50 total including taxes and fees) for one way non-stops LAX to MIA. (through January and February that I know of).
It used to be I could often get LAX to Ft. Myers cheaper than just to Miami, even though the flight would go through Miami, but that tends not to be true any longer -- especially in the height of season.
