In Prague you should buy the pilsner.
For something to take home - Prague is known for jewelry. There are many stores. If you travel about the old city you will find them. Be careful, we discovered the hard way that some of them are not very trustworthy.

i didnt see any 'musthaves ' in prague but then again i do most of my shopping locally - amazing what you think you cant buy at home only to find it right in your local mall lol
but as people here have pointed out- the shopkeepers arent very trustworthy...so do know the prices and count the change...
the face of tourism in prague is not something i remember with any great fondness
lots of rough folk doing business in the touristy bits of town
i did buy some shawls only to find that they come from asia anyway and couldve been bought at any of those little stalls we have here in our own malls

The "Rodeo Drive" of Prague is Pariska Street. Here you will find Louis Vuitton, Dunhill, and Armani. There are a couple of jewellery stores on this street that sell amber and garnets.
For expensive crystal, head to Moser on Na Prikope. (Is that Ivana Trump putting the salesgirl through her paces?)
For grooming supplies and perfume, sashay shontay over to the Dr. Botanicus behind Tyne Church in Old Town Square. You will leave smelling like bitter orange, sweetie darling.
The gift shop at the Mucha Museum has glorious stationary and postcards.

A few odd ides
Cards, lots of shops have lovely cards with nice scenes, hand made paper and good prices
They make knives which pupport to be damascene but are made out of hammered wire. Surprisingly nice but expensive
Local beer ( a few micro brewaries in restaurants) try the dark beer (very light to british standards) but interesting
