Cheapest Options
I have family in Ontario and Calgary. My husband and I are planning a trip to Canada next summer between 5 and 6 weeks long. (From UK). We would love to visit both sets of relatives as cheaply as possible. Would it be best to fly to Toronto, stay for a while then fly to Calgary internally and home from Calgary, or fly to Toronto to start then a return flight to Calgary and back home from Toronto or are there other options that we should consider? Any suggestions welcome!

You should fly to Toronto, then to Calgary and home directly from Calgary.
You will certainly save time and possibly money by doing so. Because the overseas flights use the great circle route, the flight time from Calgary to London is about 9 hours, compared to 7 or so from Toronto to London. Flying time between Calgary and Toronto is about 4 hours, so considering transit time in Toronto, your total time going from Calgary to Toronto to London would be about 12 hours or so.

You are coming for 5-6 weeks so you can do whatever is cheapest. That might be a LHR (I presume?) - YYZ return flight with a separate YYZ - YYC trip sandwiched in between.
Using July 8 - Aug 19 as dates with a the last two weeks in Calgary;
I can see a LHR - YYZ - YYC - LHR circle route for about $1724 CAN.
An LHR - YYZ return can be had for about $1070 and a YYZ - YYC return for about $625.
So, it is showing almost the exact same price, in total. If that stays the situation, you would probably not want to have to come back through Toronto from Calgary. If it gets cheaper, you need to think about how much of a difference would entice you to do the sandwich.
