Wow that's a bunch of time! Stop off at Lake Tahoe for a good long visit.
Visit Hope Valley.
Hang out for a day around Bridgeport. If the road is available go see Bodie.
Keep an eye on the opening day of Tioga Pass road and if it opens go to Yosemite.
Spend some time in Mammoth seeing all the sights.

You do know that Reno to Bishop is only about a 4 hour drive, don't you?
The area just south of Reno for about 50 miles is farm and ranch land with a few small cities along the way. I would guess, if you were interested, you could contact an agricultural group in Carson City or Minden and arrange a ranch visit or two.
Carson City is Nevada's capitol and there are a few places there to visit. Only a short detour away, Virginia City is an old silver mining town that is a combination of historic and touristy. In the last half of the 1800's, it was one of the richest silver mining districts that ever existed.
As Suzie mentioned, Lake Tahoe is only a minor detour from the direct route and has lots of interest. On the California side, it's in the heart of the High Sierra mountains. There will still probably a fair amount of snow in May but the roads should be good except during and immediately following a storm. On the Nevada side, there's a fair amount of entertainment and gaming if you enjoy that sort of thing.
Further south, Bishop is also on the east side of the High Sierras and there is quite a lot to see and do in the area.

If you go to Tahoe and then Hope Valley you can head down 50 for ahwile to El Dorado and Amador Counties for some wine tasting. Amador has some of the best zins! Then head back toward 395 if Tioga is closed. If it's not then head to Yosemite by going through the gold rush towns along 49. Take 120 into the Park and then you can exit via Tioga Pass road visit Mono Lake, Mammoth, Rock Creek before arriving in Bishop. Even in Bishop there are many day trips you could do. Go see the Bristle Cone Pine forest if the road is open. Clearly where you go will depend on how the snow has melted.

Two weeks is a lot of time. Definitely go to Lake Tahoe. In Bridgeport take Twin Lakes rd. up to Annetts Mono Village. It's a 13 mile drive to beautiful Twin Lakes and theres a campground and fishing resort. Record Brown Trout!! Also on that road is Hunewill ranch that I believe you can visit. Out of Bridgport (south on 395) is the road to Bodie. Bodie is a Historic Ghost town, but the road may not be open. See Mono Lake and then detour off into Yosemite. Mammoth Lakes area is somewhere I haven't gotten to explore yet. Sounds like a great trip. I love the eastern Sierras!!

Thanks everyone,I know Reno is close to Bishop.But We were thinking of the east of that route and calling in to kings canyon and Death valley,going down from Ely.Sorry I didn't explain myself to well.Just though there might be little places that they dont tell you about in the tour guides.We love the old fashioned diners where working people go in to for breakfast and lunch,we love to talk to locals.We are acually going to the World Mule show at Bishop can't wait.Thats why we are coming over to the US.Best of luck to you all.

Mules Days in Bishop. Fun! Fun! Fun! Are you cattle or sheep ranchers? You may want to do more internet research, such as Chamber of Commerce or tourism sites for Nevada.
Do you want to visit any ranches? The Nevada Farm Bureau may be able to set something up.
