According to hopstop.com

Take the E train past 42nd street to the 50th street stop.

Get out near W 51st and 8th avenue and walk west on 51st street to 319.

Complete directions here

Yes, but you can also take the C train if one of those comes first. You will probably see A-C-E trains together at Penn Station, but don't take the A train, it doesn't stop at 50th. Make sure you are looking at and get on the UPTOWN direction (not Downtown). When you get off the train at 50th, you can go to the far end (the direction the train is traveling) and you'll come out at 51st. You need to cross 8th Avenue and Maria Pia will be straight ahead on 51st on the left hand side.

My yes was to gb944, but I'd probably walk too if I had the time. It would take maybe 20 to 25 minutes at most if the sidewalks are crowded, going right up 8th Avenue then turn left at 51st.

Suggest you go to the mta website and download the interactive subway map. It will help a lot to have an overview in getting around the city. (Hopstop is great for one place to another, but it doesn;t provide overall perspective.)

You can also take a bus. WHat day of the week, time of day are you talking about?
Exit Penn Station at the 8th avenue side, and look for the stop for the M20 bus, which goes up 8th avenue and stops every 2 or 3 blocks.
(Subways don't always make the most sense. Sometimes by the time you get in and out of a subway station, wait for a train, possibly switch lines, and then walk a few blocks, you could have walked the whole way--or taken a bus--in much less time)
