No iron clad rule but across the board, no.
In a city dwellings, say downtown Atlanta, hotels are cheaper on the the weekend because the business travelers leave.
However, out in Reynolds Plantation, weekends are more expensive because that is where people go to escape.

From what I've experienced, the airlines are lower if you fly mid-week, vs weekends to places like Hawaii, so your flight would probably be less expensive flying mid-week. But hotels are a different story - most raise their rates for Friday and Saturday, but have lower rates during the week (Sun-Thurs). Sometimes getting a weekly rate will help. Without knowing where you're talking about going it's kind of hard to help.
