The most straitforward way would be to rent a car, and, for even two people, probably the least expensive.
If you don't want to rent a car, then you could take the R1 line to the Market East Station, walk the two blocks to the Philly Greyhound Terminal, then take the 551 NJTransit bus to Atlantic City.

For the sake of completeness:

I use Airport Chariot car service. I've been using them for a few years and they always show up & the driver's are nice. It costs about $90 for the two of us, which to me seems a lot cheaper than the other companies I originally called especially since it is an hour drive to Philadelphia to Atlantic City. The number there is 609-748-3506 and they web address is www.airportchariot.com . Atlantic City just isn't the place where a rental car is needed.

Definitely use a car service, and make it A&C Transport, they rock. Much better than one of these 'discount' ones, and at the same price. Try 'em at www.actransportonline.com
