Best and cheapest aren't the same. A taxi will probably cost about $40-45 plus tip. Bus service to Grand Central is $15 per person. There are a number of companies that will take you by limo or shuttle but prices tend to be what you'd pay for a cab. If you have little luggage, want top save a few $$ and don't mind shlepping stuff around, you could get a cab or local bus and go to Penn Station in downtown Newark, then take NJ Transit train to NY's Penn Station. Anything but the direct cab route is less than convenient since you must transfer from bus to cab or train to subway. I'm not certain whether the taxis charge by the person or by the trip - perhaps someone else can clarify.

I would recommend the SUPER SHUTTLE. Go the ground transporation desk at the airport and just tell them where you want to go. The shuttle will pick you up and drop you off right at your door. It's about $20 - and you won't have to deal with the subway w/your bags. Sometimes, you may have to wait for the shuttle, but not very long. It's a great way to go!!!
