I have only stayed at the Hilton Key West, but I would definitely recommend it. Rooms were nice, great location and a free shuttle to their private island (Sunset Key). They get my vote!

I vote for the Hyatt! In the Pier House, most rooms overlook the parking lot, and the Casa Marina is too far from the center of activity. I haven't stayed at the Hilton, but it gets VERY mixed reviews.

Go for the Hilton. Right on the pier and agree, the private island is great. You have to go early (around 10, so not too early) because they shut it down if it gets too crowded. We spent every day over there as "too crowded" on Sunset Key was way less crowded than anywhere else on Key West. Next year we're going back an booking a cottage on the island.

How about a B&B? The island has so many to choose from. If you want a hotel go with the Hyatt but a B&B would be my first vote.
