It takes about 5 1/2 hours but it really depends on the time of day and year.
There is also a ferry in Fort Myers that will take you into Key West in about 3 hours but the weather is always a factor with it and it can be canceled at the last minute due to bad weather. I do not suggest flying into Miami. Give Fort Lauderdale Airport a look. It is much easier to get in and out of quickly and many times, because it is so much smaller then Miami, actually makes the drive quicker to Key West because of getting on the road faster.

We often travel to the keys from the Philadelphia/Newark area. We use Fort Lauderdale (FLL) or West Palm Beach (PBI) and drive from there. From Fort Lauderdale, figure a little over three hours to Key West using the Florida Turnpike to Route 1. True, Miami is closer but the congestion there makes the trip longer.

We will skip Fort Myers all together. The only reason for consideration is non stop flights on Midwest Airlines from Kansas City. Originally, we wanted to fly into FLL non stop on Midwest, but the fare is $700 for our travel weekend. We can get into FLL or MIA with 1 stop for $225. I did check flying into Key West and it was around $450. Thanks for the West Palm Beach idea.
