There are lots of interesting things to see.
I presume you mean things outside the D.C. area that would make an interesting stop to break up the drive.
Without knowing your interests, here are three that we've enjoyed:
1. If coming south on the expressway (I-95), Richmond and/or Petersburg, VA both have a lot of Civil War related stuff. They may be a little too close to Baltimore, however.
2. If taking the "scenic route" (US 17), the Outer Banks or Wilmington, N.C. are both worth a visit.

17 has a ton o'stuff. New Bern, Beaufort, Wilmington, all in NC; Myrtle Beach, Pawley's Island, Georgetown, Charleston, another Beaufort, and a bunch of islands and plantations.
95? Fredericksburg has lots of history, Richmond has history, art, great restaurants. Petersburg has Civil War. Off your path is Williamsburg, the James River Plantations, Jamestown.
Below that, not much.

How much time can you take. There is lots to see and do if you use route 17. Even more if you drive from Baltimore to Chincoteague/Assateague (about 3 hours) and start from there. A couple of years ago I drove from Charleston to Columbia (near B'more) via the Chesapeake tunnel-bridge and Chincoteague and took a week +.
