We stayed at the Locanda Orseolo (www.locandaorseolo.com) last March with our kids age 9 and 12 (at the time). I believe they have a quad room, but we had two doubles just across the hall from each other which worked out great.
The hosts were wonderful with the kids. We would come back in the afternoon and rest and my 9 year old son would go downstairs and talk basketball with Matteo, and they would also let him put away and hand out the keys as guests came and went. Plus, what kid wouldn't enjoy a chocolate crepe for breakfast!?!

A few years ago we stayed in a quad at Hotel Flora. It was 1 large room with a double and 2 single beds, and 1 1/2 bathrooms. It's a very charming little hotel, and a quiet convenient location near St. Mark's.
I found when looking for accommodations for 4, it's best to email the hotels directly that you are interested in. Many only have a handful of quads/family rooms and don't list them on their websites or the booking sites.

A few years ago we stayed at the Best Western Hotel Binsanzio, just a short five-minute walk from St. Mark's Square. The room had 3 beds, a large double plus 2 twins, and even a small terrace which the kids especially liked (a little separation from mom & dad at times!). Only one bathroom however.
Some apartments may even be available for 3 nights. Check sleepinitaly or other apartment rental agencies.
