Tuscany from Florence
I too, along with my wife are planning a trip to Italy, spending 3 nights in Florence and are looking to do a day-trip (possibly an overnight) to the Tuscany wine-region.
Wondering if anyone can suggest the best way to get there via public transit, as we will not be renting a car.
Any suggestions of specific places to go in Tuscany?

First of all, Florence is in Tuscany.
Depending on the actual cities you want to visit, most of us recommend the trains to get around. Very efficient and cheap. Have you done much research yet to start narrowing down which cities you may be interested in, then let us help with the final few choices? You should do a search on this forum as the Tuscany region has many beautiful cities to offer.

i believe we will be arriving from rome via the train into stazione s.m. novelle and staying in the center of town...looking to make a day trip via public transit/rail to "wine country"...looking for suggestions of wineries/tours available by rail or any good package tours via bus possibly.

Do you want to tour a winery or taste wines?
If the latter, you can visit
Enoteca Italiana Siena, in Siena by SITA bus.
If you google <wine tasting montepulciano> you will find several firms that take you on tours.
