You could take a day out of Strasbourg I think - one full day was enough for me (and it is a wondrous city but compact) and add a day to Salzburg to do a trip into the Austrian Lakes District just to the east - a k a The Sound of Music Country - by postal bus just over an hour to St Wolfgang - take boats on the idyllic lake or a steam train up a mountain, etc.

OTOH with another day in Strasbourg you could easily day trip by train or car to nearby Colmar and the Alscae wine region - Colmar is a cute town that makes a neat day out and with car you can putz along the Alsace Wine Road and hit a few wine houses and cute wine villages:

My daughter and I went to Karlsruhe to rent bikes, and ended up staying one night coming and going. I'm not exactly sure what there is to see there, not that it is a terrible place. Based on what little I know I certainly would not plan four nights there.
I would say your time there is better spent elsewhere. And I agree with the others that Trier is worth the visit.
