There is a quick change in Karlsruhe or Offenburg to get from Strasbourg to Frankfurt by train. There is a train about every 2 hours.

The train from Strasbourg to Frankfurt(Main) Hbf takes 2:08 or 2:16 with one change either in Offenburg or in Karlsruhe. See www.bahn.de

I have been to both Colmar and Strasbourg and both are lovely and worth a visit. I agree with NanBug; for an overnight stay Strasbourg has more to offer. Be sure to see the mechanical clock in the cathedral. Bon Voyage!
