If you are arriving in Amsterdam, look at Transavia flights from Amsterdam to Pisa. They can be quite cheap if you book in advance. That's how my mother flew from Portland to Europe (she also wanted to fly non-stop). If you fly into Pisa, it is easy to get to le Cinque Terre by train, and even if you rent a car elsewhere, it is best not have one in le Cinque Terre. One way to do your trip might be fly to Pisa, see le CInque Terre, rent a car after, visit small towns, drop of car at Florence airport, see Florence, after that take train to Rome, after seeing Rome take train to Venice, fly back to Amsterdam from there (I don't know if that is Transavia too, but I know there are flights).
