Both are excellent Paradores, but we liked the Parador de Jarandilla de la Vera just that much better. The location is nice and quite, the room was very comfortable and it's the perfect place to relax along the way. There is also a swimming pool you can use during the summer months.
We didn’t stay in the Parador de Trujillo, but we did stop for lunch to check it out. It’s a little different, a little more sparse, but still comfortable.
If you need any more information, you may want to contact the Tourist office of Spain. If you want to book, then I suggest booking your stay through either Petrabax, or Totally Spain. Both of these companies deal with the Paradores and are good companies to work with.

Since you'll be driving between Seville and Madrid, be sure to pick up the Michelin Regional Map #571 which covers the area between Cáceres and Madrid. If Jarandilla is not available, then I suggest that you check into the Parador in Oropesa. Be sure to ask for one of the 4 original rooms in the Parador. They are outstanding, as is the Parador. The restaurant there serves an excellent dinner. The only problem is that it is a very popular weekend destination for people from Madrid and may be booked months ahead of time.

a stop at oropesa is well worth a detour.
it is one of my favorite, and quite an impacting castle when you first see it.
we use paradors for coffee breaks when we can on our trips if we aren't staying at them.
never a disappointment.
enjoy trujillo and its history. i can't remember how many of the adventurers off to find new lands ( the americas)came from there..
