Milan-for one afternoon
I will have a seven hour layover at Linate on October 4, 2010. Can I go from Linate, at 10 in the morning, to the Last Supper painting, with a taxi or guide and get back to the airport to fly out at 17:10? I have been to Milan twice, but have never been able to see the Last Supper. Do you think that this is possible and can you suggest a guide with tickets.

Will you have your luggage? It's possible that the church will not hold your luggage for you while you view the fresco. Check. They certainly will not let you inside with it.
If you don't have luggage, you need to make a reservation to see the Last Supper between noon and 1pm. You must reserve your ticket (everybody must) and you must pick it up 30 minutes before your reserved time. You are only allowed something like 20 minutes inside to view the fresco. You don't need a guide, although you can rent an audio guide at the church.
After you see the Last Supper, probably the fastest way to get back to the airport is to walk to Cadorna Station (10 minutes) and get a taxi to take you back to Linate. You should plan to be back at Linate by 3pm. I would make sure you are on your way by 2pm from the center of Milan.

Hopefully, your luggage will be checked through. If not, they do let you bring it inside. Due to bad advice about availability of luggage storage at Cadorna Station last year I had a large rolling bag and hand luggage. They said no problem taking it in. There were two other people in our group of 15 with luggage.

We took the luggage with us the entire way in and out. I was surprised also, one of us was prepared to miss it and stay with the luggage.
The groups are so small going through. I just parked the bags by the door that you exit and could move around and look with no problem.

I would still check to see if there has been a change in policy -- not because I disbelieve lynnaustin but because sometimes these things change. That said, if there was a change, I would probably take my luggage and ask a nearby bar or store owner to let me tuck it inside their door. They'd probably say yes, and I'd make a purchase as thanks.

Do not risk taking your luggage.
