I did it in the opposite direction two years ago on an overnight train. Too easy, and very relaxing.
You might find a bus which is cheaper but not nearly as comfortable. Try the websites of Eurolines and Ecolines for bus service.

Dresden to Prague is a 19-Euro train ticket each way, for a 2-hour ride. I can't see doing this as an overnight train nowadays. To get this price, you likely should buy your ticket well in advance at www.bahn.de .

My goof. You are right WillTravel. I was on a night train but got off in Dresden. The trip was a little less than 3 hours. The cost was €19.30 in 2006. It was an open ticket and I bought it on the afternoon of the day I departed.

I've done this route numerous times. Trains roughly every 2 hrs, and easy 2-2 1/2 hr ride.
The trains from Dresden arrive at Holesovice (sp) station. This is on the north side of town. It's a quick (10-15min) subway ride on the subway (red line) to the city center.
