Personally, I think your trip is too rushed. You have only 11 nights on the ground and you are visiting 5 destinations (including the day trip to Toledo). I love Barcelona but it is the outlier here; I would drop Barcelona if it were my trip. You only have 2 nights in Seville and 1 night in Granada. I would spend 4 nights in Seville and 2 nights in Granada. Since you will only see your son who is studying in Madrid the first week, I think it makes more sense to spend the majority of your time in Madrid that week.
This is a possible itinerary:
Madrid (4 nights including the day trip to Toledo) - I usually recommend spending at least one night in Toledo but that might be too much rushing around for the amount of time you have.
Seville (4 nights) - maybe include a day trip to Cordoba
Granada (2 nights)
Madrid (one night in Madrid - your last night)
I would take trains or the ALSA bus to each destination. No need for a car on this trip.
These are the hotels we stayed at. You can check them out to see if you like them. You didn't mention your budget.
Madrid - Mercure Madrid Centro - near the Prado and Reina Sofia
Granada - El Ladron D'Agua, Carrera del Darro - perfect location, at the bottom of the Albayzin. Parallels the Rio Darro. Few minutes walk from Plaza Nueza.
Seville - Apartamentos Suites Santa Cruz, Calle Pimienta 4 - located in a pedestrian only area. We had a one-bedroom apartment. A few minutes walk from the Cathedral and Alcazar.

With the time you have available, I think I'd go either north of Madrid or south but probably not both. You could also consider places like Valencia, Salamanca, Avila, Segovia, etc. if you wanted to stay more horizontal/central.

Look at travel time, including time getting to train stations from hotels and to hotels from stations. At best, each travel day is going to use up about 1/2 a day, which is really important with short winter days.
With ten days on the ground, you have only six full days without some kind of travel and some of those travel days are longish.
You are locked into RT Madrid, so I agree with others. Drop Barcelona.
Definitely add two nights to Seville.
Add Cordoba as a day trip from Seville. You could do a night there, but easier to add the night to Seville and make Cordoba a day trip.
Add a night to Granada.
It could be very cold in Madrid and Granada, just a heads up.

I have just returned from another nine nights in Madrid ( only) , but
some travellers prefer a quick. “ taste “ of a city and fast paced itinerary.
You have been given good advice here, but depending on your energy and interest , anything is possible.

Here is your itinerary with added commentary:
Wednesday, November 16, 2022 Arrive Madrid 12:25 pm 1st night, see various monuments if time after you arrive at your hotel.
Thursday, November 17, 2022 Madrid - Toledo: yes a Thursday is a good day to see Toledo. Be careful about going on a Monday or Tuesday because some places are closed on Mondays or Tuesdays. 2nd night
Friday, November 18, 2022 Madrid: 3rd night in Madrid. In March 2022 when I went to Spain I had 13 nights on the ground, 1 night on the plane on the way there, 5 nights in Madrid at the end of my trip. I suggest putting all your nights in Madrid at the end of your trip. Go to Seville on a train (or bus) the day your plane arrives.
Saturday, November 19, 2022 Madrid to Seville : 4th night, 1st night in Seville.
Sunday, November 20, 2022 Seville. 5th night, 2nd night in seville. I spend 4 nights in Seville. You really only are giving yourself one full day here. Are you sure you are only interested in seeing the Alcazar or whatever else you might cram in?
Monday, November 21, 2022 Seville to Granada : I also flew round trip to and from Madrid, but I went to Seville the day my plane arrived. 6th night, only one night in Granada?? I spent 2 nights in Granada, after Seville. With only the one night in Granada, you would have to rush to pick your luggage up from your hotel and then to the train station or bus station or airport for an evening flight or train or bus.
Tuesday, November 22, 2022 Granada to Barcelona. 7th night. I skipped Barcelona. After Granada I went to Cordoba.
Wednesday, November 23, 2022 Barcelona 8th night
Thursday, November 24, 2022 Barcelona 9th night, 3rd night in Barcelona.
Friday, November 25, 2022 Barcelona to Madrid 10th night
Saturday, November 26,Madrid. 11th night
Sunday, November 27 departing Madrid
Have you considered seeing Madrid or Barcelona but not both; or seeing both but skipping southern spain; or just the 3 cities of Madrid, Barcelona, and Seville? Check the times you can leave Granada and make sure one night there will be enough or see if you need 2 nights in Granada.
