It is feasible, but I would urge you not to do it. You will do a whirlwind tour through the highlights, and if delayed (as often happens in Italy) would feel frustrated.

Have you considered a trip to Orvieto? It takes an hour, Costs 11 or 15E one way, is a much smaller town that you can see liesurly, and you get to ride the funiculare.

I actually did the day trip from Rome to Florence when my fiancè came to visit me once. We took the 7.30 train from Rome, arrived in Florence at 9 am, had breakfast, then strolled from the Station Santa Maria Novella to the church (s. Maria in Fiore and the Battistero) and then from there the Uffizi museum. Lunch at Ristorante ai 4 amici and then shopping in the city centre along Ponte Vecchio. We took the late afternoon ES train with the good feeling of having spent a wonderful day. Of course you CANT see a lot of things but if you try to concentrate the basic things you should spend a lovely day in Florence.

It's nice to fill in a day with a trip to Florence. The Duomo, the Piazzas, churches, some shopping and a nice lunch are all do-able. A museum (e.g. Uffizi, Accademia) might be a stretch. After our daytrip, we longed for and made sure we had a longer stay on our next trip - which we did and we just kept coming back.
BTW, the cost as per trenitalia.com is around $34/$49 (2nd/1st class) one way.

The last Eurostar train back to Rome from Florence leaves at 22:54 (arrival Rome half past midnight), so you COULD spend nearly 14 hours on the "day trip" to Florence, if you have the energy of Superman!!!!!!!!!!!!

Back in the day when they were practically giving high-fashion items away in Florence, I would day-trip from Rome just to go shopping. Shopping is still great in Florence (few bargains) but my need (desire) to own so much stuff has diminished. I still get tempted to day trip and I won't hesitate when the moment strikes.
