I can't remember precisely how long the drive is - but it takes you through some gorgeous countryside.
As far as must-dos - Porto Ercole is pretty small and touristy activities are sort of few and far between in the city itself. It is extremely picturesque and would be a good relaxation point.
It is also a decent jumping off point to visit other cities in Tuscany...not sure if you are wanting to do that or not.
