I think St. Mark, mentioned by Annhig is another great place for a short time in Rome>>
Bvl - i think that it's obvious, but in case it isn't, I think that you mean the monastery of San Marco in Florence, not Rome. but i agree that it's a very good place to go to enjoy some of the most extraordinary art in Florence in relative peace and quiet.

In one day forget seeing anything particular inside of any place and just wander - if the door of a church is open and then is no throngs to get inside - ubiquitous at the Duomo in Florence - then wander in - the churches, like the roads, less traveled are often IME the nicest surprises.

In 2006 I went to Rome for a week with my then 15yo daughter, and we did a day trip to Florence.
I posted a trip report, which focussed on what gave my daughter those WOW moments, when she came across something amazing which she hadn't been expecting. Here's the Florence excerpt...
<<We did a day trip to Florence which takes 1 1/2hours on Eurostar, round trip 117 euros. We both really enjoyed this, saw David (WOW) at the Academia (pre-booked), Medici Chapel (WOW) and the Baptistery (WOW). We shopped in the markets, lunched, wandered around, and had a really nice (and good value) meal at Maso on the corner of Via Palazzuolo and Via Maso Finiguerra on our way back to the station. We arrived back in Rome around 10pm.>>
Although it rained all that day, it gave us a nice taster of Florence, and a few years later we returned for 3 days. Obviously there is a great deal to see and do, but pick your priorities, get an early train, and you can still get a good overview of the city before getting a train back around 8.30pm.
