I don't know if there is a bus line, but I was taking an IC train a couple of weeks ago from Rome to Salerno. A woman was continuing on to Sicily. It was a nine hour train ride for her. I can't even imagine being on a bus. You may want to reconsider!

Italian trains are so good and so cheap that there are very few long-distance bus services, except on routes where the trains don't go. If you are travelling to Sicily, there are through trains which cross the Straits of Messina by ferry - you can get on a train in Roma and alight in Palermo, Siracusa, Agrigento or numerous other places in Sicily; on daytime trains, most people get off the train during the ferry crossing to get refreshments and stretch their legs, but on overnight trains, you sleep through the ferry crossing.
